ワシントンDCの地元の有名なデパートで勤めていた母と掃除人の父の元に生まれたタラジ・P・ヘンソン。大学をジェシー・ジャクソン等が卒業している歴史的な黒人大学ノースカロライナ農業・工業大学に進むも最初で躓き、すぐに名門黒人大学ハワード大学に移籍した。朝はペンタゴンで働き、夜はディナークルーズのバイトで大学のお金を稼いで、ハワード大学をシアターアーツの学位を取得して1997年に卒業している。

卒業と同時に人気TVシリーズ「ER」等にゲスト出演するようになり、1998年のインディペンデント作品「Streetwise」で映画デビュー。その後もコンスタントにTVシリーズのゲストや映画に出演し、2001年のジョン・シングルトン監督の「Baby Boy / サウスセントラルＬＡ」にて主役のタイリースの恋人を演じて注目されるようになった。

そして2005年の「Hustle & Flow / ハッスル＆フロウ」では、主役のテレンス・ハワードの恋人の一人を演じ、劇中でも歌っていた「It's Hard Out Here for a Pimp」をアカデミー賞の大舞台で歌った。2007年にはドン・チードルとキウェテル・イジョフォーと共に「Talk To Me」に出演。数々の賞にもノミネートされて、ファンの間では好評を得た。

2008年には主役のブラット・ピットの母親役として「The Curious Case of Benjamin Button / ベンジャミン・バトン　数奇な人生」に出演し、スクリーン・アクターズ協会賞や放送映画批評家協会賞等にもノミネートされ注目を集めている。
